The effect of Haemonchus contortus infection in sheep fed with a moderate and high protein content diet was evaluated in two breeds of sheep. Forty-eight Ile de France and Santa Ines lambs were maintained indoors since birth, in worm-free conditions. The lambs were allocated after weaning in four groups of six animals per breed, which were either infected or remain uninfected and given access to either a moderately or highly metabolizable protein diet. The moderately and highly metabolizable protein diets were calculated to supply 75 and 129 g metabolizable protein per kg of dry matter (MP/kg DM), respectively. The infection consisted of a trickle infection with 300 infective larvae, three times a week, for 12 weeks. Significant differences were observed for mast cell, globule leukocyte and eosinophil counts in the abomasal mucosa of the infected groups compared to the control of both breeds (P<0.05), regardless of the diet supplied. Significantly higher IgA anti-L5 antibody was detected in the infected Santa Ines groups than in the infected Ile de France groups (P<0.05). Increased metabolizable protein supply resulted in larger body weight gain and higher packed cell volumes for both breeds (P<0.05). Both breeds showed an increased ability to withstand the pathophysiological effects of H. contortus infection when given access to the highly metabolizable protein diet. However, increased metabolizable protein supply resulted in reduced worm burdens in Santa Ines lambs but not in the Ile de France lambs (P<0.05). The present results show that the increase in protein content in growing lamb diets may benefit resistance and resilience to gastrointestinal parasites but that these benefits may vary among breeds.  相似文献

This study was carried out to evaluate the immune response and the impact of Trichostrongylus colubriformis infections on the performance of growing Santa Ines lambs. Thirty male lambs, 3-4 months of age, were maintained in individual pens and restrictively randomised by weight into 3 treatment groups: (1) infected group, artificially infected with 2500 T. colubriformis larvae, three times a week, for 13 weeks, and fed ad libitum; (2) Pair-Fed Group, non-infected and fed with the same amount of food consumed by the infected animal of the same class on the previous day; and (3) control group, non-infected and fed ad libitum. Refused feed was weighed daily to assess the food intake of each lamb. Animals were weighed weekly and blood and fecal samples were collected. At the end of the trial, all lambs were euthanized to determine worm burden and collect intestinal tissues and mucus samples for histological and immunological analysis. The infected group presented eosinophilia, increased number of inflammatory cells in the mucosa, in addition to an increased production of specific immunoglobulins against T. colubriformis, which partially prevented the establishment of infective larvae. As a consequence of parasitism, the infected lambs presented reduced serum albumin concentrations and demonstrated severe small intestine lesions, such as villous atrophy and epithelial erosion, which impaired the digestion and absorption of nutrients, causing a significant loss in performance. The infected group presented a 37% reduction in daily weight gain (107.26 ± 10.8 g/day), when compared with the control group (171.07 ± 7.15 g/day). The infected lambs also demonstrated the worst food conversion, i.e., each animal needed to consume on average 10.05 ± 0.52 kg of food to gain 1 kg live weight. The voluntary hay intake depression in infected animals was small, and statistical difference (P<0.01) was seen only on two occasions (ninth and 12th weeks), in comparison with the control group. In conclusion, Santa Ines lambs infected with T. colubriformis demonstrated a reduction in performance, caused by the damages produced by the adult nematodes in intestinal mucosa, and also by the immunopathological changes associated with the infection.  相似文献

The most challenging obstacles to testing products for their anthelmintic activity are: (1) establishing a suitable nematode in vitro assay that can evaluate potential product use against a parasitic nematode of interest and (2) preparation of extracts that can be redissolved in solvents that are miscible in the test medium and are at concentrations well tolerated by the nematode system used for screening. The use of parasitic nematodes as a screening system is hindered by the difficulty of keeping them alive for long periods outside their host and by the need to keep infected animals as sources of eggs or adults when needed. This method uses the free-living soil nematode Caenorhabditis elegans as a system to screen products for their potential anthelmintic effect against small ruminant gastrointestinal nematodes, including Haemonchus contortus. This modified method uses only liquid axenic medium, instead of agar plates inoculated with Escherichia coli, and two selective sieves to obtain adult nematodes. During screening, the use of either balanced salt solution (M-9) or distilled water resulted in averages of 99.7 (± 0.73)% and 96.36 (± 2.37)% motile adults, respectively. Adult worms tolerated DMSO, ethanol, methanol, and Tween 80 at 1% and 2%, while Labrasol (a bioenhancer with low toxicity to mammals) and Tween 20 were toxic to C. elegans at 1% and were avoided as solvents. The high availability, ease of culture, and rapid proliferation of C. elegans make it a useful screening system to test plant extracts and other phytochemical compounds to investigate their potential anthelmintic activity against parasitic nematodes.  相似文献

Haemonchus parasites are responsible for many losses in animal production. However, few studies are available, especially of zebu cattle. In this study, we investigated mRNA differences of immune response genes in na?ve Nellore calves infected with Haemonchus placei, relating these differences to patterns of cellular infiltrate. Calves were infected with 15,000 H. placei L3 larvae and after 7 days lymph node and abomasum tissues were collected. IL-2, IL-4, IL-8, IL-12, IL-13, IFN-γ, MCP-1, lysozyme, pepsinogen and TNF-α genes were evaluated by qPCR. Mast cells, eosinophils and globular leukocytes were counted by abomasum histology. In the infected group, IL-4, IL-13 and TNF-α were up-regulated in the abomasal lymph node. In the abomasum, IL-13 increased and TNF-α was down-regulated (p<0.05). No differences were detected for mast cells and eosinophil counts in abomasal tissue (p>0.05). We conclude that for this infection time, there was Th2 polarization but that cellular infiltrate in abomasal tissue takes longer to develop.  相似文献

The effects of nitrogen fertilizer treatment and source (prilled urea, urea supergranule, fresh azolla, rice straw or sesbania or rice straw compost and their combinations) on grain quality were studied in the 1987 crops of variety IR64 at IRRI. Although fertilizer application improved grain yield, it improved protein content only in the case of urea supergranule, azolla and rice straw. Lysine contents of brown rice protein were similar in samples with no N fertilizer and those with the highest protein content in both seasons. Fertilizer treatment regardless of source tended to decrease weight and increase translucency of brown rice in both seasons. Effects on other grain properties were not consistent in both seasons. Season affected more grain properties than fertilizer treatment did, particularly translucency which was higher in the dry season than in the wet season.  相似文献

Receiver functions derived from teleseismic body waves recorded by ocean-bottom seismometers on the southern East Pacific Rise reveal shear waves converted from compressional waves at the mantle discontinuities near 410- and 660-kilometer depth. The thickness of the mantle transition zone between the two discontinuities is normal relative to the global average and indicates that upwelling beneath the southern East Pacific Rise is not associated with an excess temperature in the mantle transition zone.  相似文献

This trial was carried out in Piracicaba, São Paulo State, Brazil, to comparatively evaluate the degree of resistance to naturally acquired gastrointestinal nematode infections in sheep of the following genetic groups: purebred Santa Ines (SI), SI crossbred with Dorper (DO × SI), Ile de France (IF × SI), Suffolk (SU × SI), and Texel (TE × SI). Fifteen ewes from each group were raised indoors until 12 months of age. At this age, they were moved to pasture that was naturally contaminated by nematode infective larvae and were evaluated from December to May, 2007. Rainfall ranged from 267 mm in January to 37 mm in April. Maximum and minimum mean temperatures ranged from 32.5 °C to 19.0 °C in March and from 25.9 °C to 12.8 °C in May. There was an increase in the mean number of eggs per gram of feces (EPG) after animals were placed on pasture with significant difference between the SI (80 EPG) and IF × SI (347 EPG) groups in January; and the DO × SI (386 EPG) and TE × SI (258 EPG) groups in May. The highest mean fecal egg count (FEC), 2073 EPG, was recorded for the TE × SI group in February. All groups showed a progressive reduction in body weight throughout the experiment of 12.0% (TE × SI) to 15.9% (SU × SI). In general, the animals with the highest FEC presented the lowest packed cell volumes (PCV); the highest correlation coefficient between FEC × PCV occurred in the SU × SI sheep in January (r = −0.70; P < 0.01). Similarly, there was an inverse relationship between FEC and blood eosinophil values, with the highest correlation coefficient in the TE × SI sheep in February (r = −0.64; P < 0.05). Immunoglobulin G (IgG) levels against Haemonchus contortus antigens increased in all groups as a result of the exposure to parasites and remained relatively constant until the end of the study, with the exceptions of SU × SI and TE × SI, which showed a rise in IgG levels during the last sampling that coincided with a reduction in mean FEC. In conclusion, crossbreeding Santa Ines sheep with any of the breeds evaluated can result in a production increase and the maintenance of a satisfactory degree of infection resistance, especially against H. contortus and Trichostrongylus colubriformis, the major nematodes detected in this flock.  相似文献

Infection with the gastrointestinal nematode Haemonchus contortus causes considerable losses in the sheep industry. In this study, we evaluated the effect that climate has on third-stage larvae (L3) of H. contortus in terms of their migration from sheep feces to Brachiaria decumbens grass, as well as their distribution among the forage plants. Fecal samples containing H. contortus L3 was deposited on the soil among the herbage at an initial height of 30 cm. Sample collection began 24h after contamination and was performed on alternate days over 13 days. The L3 were recovered and quantified in three strata (heights) of grass (0-10 cm, 10-20 cm and >20 cm) as well as in the remaining feces and a superficial layer of soil, collected from beneath the feces. In order to obtain results under different environmental conditions, fecal samples containing H. contortus L3 were deposited on pasture in January (summer), in April (autumn), and July (winter). In all of the periods, the L3 were able to migrate from the feces to the herbage. However, rains, accompanied by high relative humidity and high temperatures, apparently favored migration. The highest L3 recovery rate in the pasture was in the summer observation period, which had the highest number of days with measurable precipitation, high relative humidity (>68.2%), and the highest temperatures at the soil level (minimum and maximum means of 19°C and 42°C, respectively). Under those conditions, larvae began to reach the upper stratum of the grass (>20 cm) by 24h after the deposition of fecal matter, the number of larvae having reached that stratum peaking at seven days after deposition. In the autumn observation period, there was no rainfall in the first five days post-contamination. During that period, high numbers of larvae were found in the fecal samples demonstrating that feces can act as a reservoir of larvae in the absence of rain. Except for two days in the summer observation period, when most of the L3 were recovered from the tops of blades of grass, L3 where located predominantly at the base of the herbage. In conclusion, rainfall favors the migration of L3 from feces to herbage. In addition, larval migration up and along blades of grass can occur relatively rapidly when the temperature is high.  相似文献

Hematophagous gastrointestinal parasites cause significant economic losses in small ruminant grazing systems. The growing reports of multi-drug resistant parasites call for intensive research on alternative treatments for anthelmintics to help small ruminants cope with these parasites. Two-month-old lambs with mean body weight (BW) of 22.5 kg were experimentally infected with a multidrug-resistant Haemonchus contortus strain. Infected animals were dosed orally with Cymbopogon schoenanthus essential oil to evaluate its anthelmintic potential. Eighteen animals were allocated into three groups of six animals, and each received one of the following treatments: Group 1 - control (10 mL of water), Group 2 - C. schoenanthus essential oil (180 mg/kg BW); and Group 3 - C. schoenanthus essential oil (360 mg/kg BW). Animals received the oil once a day for 3 consecutive days. Lambs were evaluated clinically for blood biochemistry before, at 1, 5, 10, 15 and 20 days after treatment, and then were euthanized to assess the total worm burden. No statistically significant reduction in fecal egg count, packed cell volume or total worm count was observed after treatments. Also, no statistical difference among group means for blood levels of urea, creatinine, albumin, alkaline phosphatase, aspartate aminotransferase and gamma glutamyl transferase was found. Larval development assay (LDA) and egg hatch assay (EHA) were performed from feces of treated animals at 1, 5, 10 and 15 days after essential oil administration. An inhibition in LDA was observed 1 day after the 3-day treatment in larvae from feces of animals treated with 360 mg/kg essential oil. In conclusion, the essential oil at the doses of 180 mg/kg and 360 mg/kg was safe to sheep, but failed as an anthelmintic treatment when applied to young sheep artificially infected with a multidrug-resistant H. contortus strain.  相似文献
